Construction of an EMS induced mutant library and identification of morphological characteristics in Medicago sativa

Abstract: The effects of different concentrations of EMS on seed germination and seedling growth in Medicago sativa were analysed and the morphological characters of M1 mutants identified. The final germination rate, germination index and the length of radicle and germ of M1 seeds were restrained after EMS treatment. There were 379 mutants from 1039 M1 populations showing leaf (but excluding leaf shape and quantity), plant height, branch number, plant type mutations. The mutant lines of leaf color, branch number, plant type and others were 215, 40, 95 and 28 respectively and the frequency was 20.69%, 3.85%, 9.14% and 2.69%. The mutant library we constructed will be a very useful genetic resource for functional genomics and genetic improvement of M. sativa.
